Background

Thermostable RNase H is a ribonuclease enzyme that can specifically hydrolyze the phosphodiester bonds of RNA that hybridizes to the DNA strand under high temperature conditions, so it can degrade RNA in the DNA/RNA hybrid strand without digesting single-stranded or double-stranded DNA. The heat-resistant RNase H and the RNase H in E. coli are homologous proteins and both have endonuclease activity, but the heat-resistant RNase H can also be active at 65 °C, which allows the enzyme to be used for high-temperature experiments. The enzyme gene was derived from the extreme thermophilic bacterium Thermus thermophilus, which was expressed and purified in the plasmid of Escherichia coli.
